Try this out
=AND(COUNTIF($L$2:$L$5000,L2)=1,ISNUMBER(L2),L2>0)
Your formula will restrict duplicate entries but not letters..
However, this formula will restrict numbers < 1, along with restriction towards entering any letters and duplicates..

=AND(L2>0,COUNTIF($L$2:$L$5000,L2)=1)
