calculating difference of two running totals and watching it in another cell

How do I calculate the difference between two 'running total' quantities and output it in another cell?
The running totals get updated every time, and their cell addresses/locations also change. So, how do I do it?

If you do =[first_cell]-[second_cell] then the cell addresses in the formula should update automatically when the cells move.
