Cell A1 = £10,000

The usual way to calculate this would be:

=SUM(A2:A4)

E.G.
A2 = £5,000
A3 = £2,500
A4 = £2,500

However, I want my colleagues to enter the numbers without automating the formula "=SUM(A2:A4)".

However, I don't want to autosum cell A1, but I want to create or use a formula that will highlight that cell A1 doesn't equal A2:A4.

Any advice on how to do this?

Something like:
Formula:
`Please Login or Register  to view this content.`

in another cell

Or would you prefer cell A1 itself to be highlighted?

You can add a conditional formatting rule for A1 that says

=A1<>SUM(A2:A4)

Did you see 6String';s suggestion above?

If you are unfamiliar with Conditional Formatting...
1. highlight the range you want to apply the conditional formatting to
2. on the home tab, styles, select CF
3. select new rule, select use formula
4. enter =A1<>SUM(A2:A4) Format Fill as needed
