I have been working on a spreadsheet and saved. Upon re-opening it I receive the following message, screen grab attached too:
Excel Cannot Open the File/the File Format or File Extinction Is Not Valid. ... Verify that the file has not been corrupted and that the file extension matches the format of the file.
i've attempted a repair, which fails. I have full permissions on the file.
Have you repaired your Office installation also?

The message just tells you that there is something wrong with the contents of the file.
You're using Office 365 so the extension is either xlsx (which it shows) or you erroneously named it xlsx while it's an older Excel version and should have been xls.
Another plausible reason is that you have a customized ribbon but that I cannot tell from the information you have provided.
Guessing and assuming won't help you.
You will have to recap what you did before this happened, what did you change, etc.
I know it's not much to go on for you but ...
If the file doesn't contain private data you can attach it as it is and maybe someone can see what's wrong.
You can also open Excel in safe mode, hold down the Ctrl of Shift key while the file is double-clicked to open

1- Copy your filename.xlsx on C drive (for quick finding)
2- Run command prompt as an administrator (for all rights)
3- Change file extension to xls using rename command "rename filename.xlsx filename.xls"
4- Open file in excel.... enjoy
