Unable to combine Date and Time

In Cell B2 Ive entered a date. In Cell B3 Start time and in B4 End time. I want to be able to combine the date and time as displayed in Cells E2 and E3.

Also is there a way of being able to do the above, autoformatted, in order to get yesterdays date and time together.

In B2 you've entered a date and time. If you just want the date, change the function from NOW() to TODAY().
