The answer is A6+S6+CS6 !
The above can't be correct, so you must be wanting some indirect function here? Perhaps
Indirect("A1")+Indirect("S6")+Indirect("CS6")

Do you mean this?
=SUM(B1:B6,C1:C6,CS1:CS6)
What do you mean by "that also contain letters"? You are going to need to offer a lot more specific detail. We are not mind-readers.
