autogenerate reference numbers

I'm looking to auto generate reference number in 1 column that are in order. I don't want to generate the number until info is entered in the column after it. anyone done this before?

I figured this one out but have another question. Is there anyway once these numbers are generated to look them in where they cannot be changed?
