I am trying to create conditional formatting in cell E7. If E7 does not equal cell B10 I would like to highlight cell E7. I saw a similar post, but could not get the solution to work for me. thanks in advance.

You can do this with a formula. Select E7 then go into conditional formatting, select New Rule, select "Use a formula to determine..." and use this formula
=E7<>B10
or more simply by selecting "Format only cells that contain" then select
Cell Values - Not Equal To - =$B$10
