How to count cells with question marks in Excel?

Instead of the question mark use Char(63) in your formula.

Or using Tims answer from above, try:

=COUNTIF(A1:A15,"*~?*")

wildcards only work as wildcards in certain scenarios, so assuming you want to count cells that are exactly equal to ? in A2:A100 you could use this formula

=SUMPRODUCT((A2:A100="?")+0)
