1) 2 column of numbers(maybe 50) on 1 worksheet

2) would like the left column to substract right column and have the output in the other worksheet.

Put a formula in the first sheet doing the calculation want need
Move (Control + x) the column with calculation to the second sheet

End(3) - same as End(xlUp)
(2) is a row offset for the last cell in the column without data.

Entire statement End(3)(2) same as End(xlUp).Offset(1)
